Finally got to the bottom of this, Macro was nice enough to send me the PAL I was missing. As it turns out putting anything in that socket stops the game from booting, including the PAL that *should?* be in there.
After a lot of googling I managed to find a couple of low res pcb pictures, and it looks like there was a version of this game that left that socket empty as I found two different pictures with that socket empty. Most of the pictures I found had it populated though. Also the issues I was having was caused by the large custom in the corner of the board, after reflowing the pins I am now able to credit the game and the sound settings save.
Thanks again to everyone that helped.
Edit: Final update (hopefully), was playing the game today and noticed some sound issues so I replaced all the caps in the sound section and sure enough a few had started to leak. Now the missing PAL in the socket doesn't stop the game from running.

It's obviously not needed but I'll leave it in there since I have it.
