Steamin' Steve: I remember there being a train station in Gun.Smoke. Maybe you could play with this one a bit, like have your guy ride on horseback and shoot bandits in the carriages? Then Steve could throw TNT at you from the engine once you reach it, after he detaches the rest of the cars. Engine would be at the top of the screen but since there's multidirectional shooting, you could maybe add a phase where you circle around and behind the engine from left to right.
Neck Snapper Nick looks a bit difficult to pull off, at least in terms of mechanics. Will he lasso you? Maybe he could attempt to pull you next to him, then if he succeeds he hangs you and it's game over. You could make it so that he does slow bullet spreads, then forces you to weave between them as you avoid the lasso. When you do get caught, you could probably mash buttons to escape.
Bison Brothers could be a top-view version of that stampede sequence from Sunset Riders. Maybe there could be a mechanic where you could possibly cause a bison to whack one of the brothers for points and extra damage? Otherwise I was tempted to think they'd be pulling Psycho Crushers on you from both sides of the screen...

Stagecoach Sam: You could chase the stagecoach, but that would be phase one, probably an entire level trying to bring it down before the time limit or something. When you do, he hops off and faces you in a duel to the death. "RULES OF THE FRONTIER, JACK!" *inappropriate metalcore music starts playing*
