Anyone know how to reach the true final boss of Caladrius?

Can I restart a stage at least? Or can I do it on any difficulty? And I can also use bombs?

Do I have to clear bonus stages as well? Don't fail them?

I think you can restart a stage, though I haven't really tested it, since, its been a while playing it.

Yes, you can play it on any difficulty, including Practice difficulty. And yes, bombs can be used.

The true final boss is Beelzebub, right? Because every 1CC run I see has Beelzebub as the final boss. Also it looks like she's the one responsible for driving King Graham to madness.

I've already beat her. Had to restart a few times to get to her, though.
