Generally people use the 5mm Midi Perler/Hama beads. I personally use perler.
I take a snapshot of what I want to do in mame, open it in paint and zoom in 800%. Basically every pixel is 1 bead, and I count how many pixels long and how wide it is before doing it to see if I have enough space and feel like I can make it.
Not exactly sure what you mean here, it's usually better to just to do the sprites in their exact size in my experience. I don't really know how to make a bigger sprite to work with without distorting things.
The pegboards you use can be arranged as you see fit, keep in mind they are 29x29, so if you need to make something that is bigger than 29x29 you might need 4 pegboards in order to fit it easily.
